Carolin Hoffmann est\u00e1 a liderar uma investiga\u00e7\u00e3o inovadora sobre a liga\u00e7\u00e3o entre a disfun\u00e7\u00e3o do sistema imunit\u00e1rio e perturba\u00e7\u00f5es da sa\u00fade mental, como a esquizofrenia.<\/p>\n\n\n\n<p>Este trabalho pioneiro explora como o sistema do complemento, essencial no combate a infe\u00e7\u00f5es e no desenvolvimento cerebral, pode contribuir para estas condi\u00e7\u00f5es. Utilizando ferramentas avan\u00e7adas como o CRISPR\/Cas9 e organoides cerebrais, a Dra. Hoffmann est\u00e1 a desbravar caminhos que poder\u00e3o transformar o diagn\u00f3stico e o tratamento destas doen\u00e7as.<\/p>\n\n\n\n<p>No entanto, para que esta investiga\u00e7\u00e3o continue a progredir, o apoio da comunidade \u00e9 essencial. Estamos a lan\u00e7ar uma campanha de angaria\u00e7\u00e3o de fundos para ajudar a financiar este projeto e acelerar descobertas que poder\u00e3o beneficiar milh\u00f5es de pessoas.<\/p>\n\n\n\n<p><strong>Doa\u00e7\u00e3o online atrav\u00e9s da plataforma PPL:<\/strong>\u00a0<a href=\"https:\/\/ppl.pt\/causas\/fatoresgeneticos\" target=\"_blank\" rel=\"noreferrer noopener\">https:\/\/ppl.pt\/causas\/fatoresgeneticos<\/a><\/p>\n\n\n\n<p><strong>Qual \u00e9 o contexto desta investiga\u00e7\u00e3o?<\/strong><\/p>\n\n\n\n<p>Os dist\u00farbios do neurodesenvolvimento, como os dist\u00farbios do espectro do autismo ou a esquizofrenia, afetam o desenvolvimento cerebral e impactam milh\u00f5es de pessoas em todo o mundo. Estes dist\u00farbios surgem de uma combina\u00e7\u00e3o de fatores gen\u00e9ticos e ambientais que alteram o funcionamento cerebral nos est\u00e1gios iniciais da vida. Pesquisas recentes destacam o envolvimento do sistema imunit\u00e1rio e de um gene pouco estudado (vamos cham\u00e1-lo de gene X) no desenvolvimento e funcionamento do c\u00e9rebro. Contudo, o papel exato deste gene nos processos de neurodesenvolvimento permanece pouco compreendido, criando a necessidade de estudos adicionais.<\/p>\n\n\n\n<p><strong>Qual \u00e9 a import\u00e2ncia deste projeto?<\/strong><\/p>\n\n\n\n<p>Este projeto visa preencher uma lacuna cr\u00edtica no nosso entendimento sobre como o gene X influencia o desenvolvimento cerebral. Os dist\u00farbios associados ao desenvolvimento neuronal t\u00eam um impacto profundo nos indiv\u00edduos, fam\u00edlias e na sociedade, mas os tratamentos eficazes continuam limitados devido ao conhecimento incompleto das suas causas subjacentes. Ao estudar a fun\u00e7\u00e3o do gene de interesse, esta pesquisa poder\u00e1 revelar novos alvos terap\u00eauticos para o tratamento destes dist\u00farbios. Al\u00e9m disso, ser\u00e1 criada uma linha celular knockout para o gene X, avan\u00e7ando nos m\u00e9todos de estudo do desenvolvimento cerebral em laborat\u00f3rio.<\/p>\n\n\n\n<p><strong>Quais s\u00e3o os objetivos do projeto?<\/strong><\/p>\n\n\n\n<p>O objetivo principal deste projeto \u00e9 compreender como a inativa\u00e7\u00e3o do gene X afeta o neurodesenvolvimento, utilizando organoides cerebrais derivados de c\u00e9lulas estaminais humanas.<\/p>\n\n\n\n<p>Iremos usar a tecnologia CRISPR\/Cas9 para inativar a fun\u00e7\u00e3o do gene numa linha de c\u00e9lulas estaminais pluripotentes induzidas humanas (iPSCs). Estas c\u00e9lulas ser\u00e3o ent\u00e3o usadas para cultivar organoides cerebrais, uma estrutura 3D semelhante ao tecido cerebral embrion\u00e1rio. Ao comparar os organoides com e sem o gene, podemos examinar o efeito do gene X nas altera\u00e7\u00f5es da composi\u00e7\u00e3o celular e matura\u00e7\u00e3o, forma\u00e7\u00e3o de camadas corticais, migra\u00e7\u00e3o neuronal e estrutura sin\u00e1ptica e dendr\u00edtica.
